Mrs. MacDonald is Molly MacDonald & James MacDonald's mother. Oddly, she isn't a rabbit like Molly and James (lacking the ears and face), but has the same hair as her daughter so the MacDonald kids probably aren't adopted. It's assumed that Molly and James's racial trait is probably inherited genetically from their father, but nobody's really sure since he's never been seen.
